Already the Workstation has 4X8GB RAM in it. Need to add an extra 32 GB in it. Can we directly add 1 32GB RAM or 4X8GB RAM or 2X16GB RAM.
Will there be any running issues if we do add 1 32GB RAM in it instead of 4X8 or 2X16 GB RAM?

The specs for your system would be found here. And I believe you stated that you are running 32 GB (4 x 8 right now and want to add another 32 GB of RAM. In that case you could go 8 x 8 GB, or 4 x 16 GB, or 2 x 32 GB of RAM. All of those configurations would give you a total of 64 GB of RAM. But considering that you have 4 x 8 GB now, I would add 4 x 8 more so that the system will run in Quad Channel and for the best compatibility.
