READYBOOST is designed to put some of the files that Windows needs when starting-up onto a fast device, such as a USB memory-stick, to make start-up take less time.
If your "C:" disk-drive is a SSD (or NVM-E), then it is the same speed as a USB memory-stick will be. Thus, there is no "speed-up" when using READYBOOST in such a configuration.
So, how old is your disk-drive? If it is more than 3 years old, it may be having minor problems, such as an automobile tire where most of the tread has worn off, making the tire a candidate for replacement.
