The printer must be connected to the Internet in order to communicate with the Instant Ink service.

The connection depends on Web Services.

Instant Ink service monitors your printer - the printer reports to the service to provide information on pages printed, printer condition, and whether it is time for the service to send additional ink cartridge(s).

You can switch off your printer for reasonable periods of time, of course.

Switching off the printer does not switch off the subscription.

When the printer is again switched on and connected to Web Services, the service and the printer will communicate the latest information for each.

In general, unless you have a very good reason for switching off the printer, it is easier on the printer to leave it "on" and let it sleep.  "Sleeping" printer will awake occasionally to perform self-maintenance and to connect (through Web Services) with the Instant Ink subscription.
