"Update Now" button is not available in File>Account Menu in MsOffice2016

If the "button" you expect is not available,

If Windows Update should be updating the software but is not doing so,

I would guess the software needs to be "repaired" or removed and reinstalled using your Office License key.

Would that work?  Unknown.
