HP Z420 work station
Microsoft Windows 10 (64-bit)

I have a HP Z420 work station that is using a ASUS graphics card (see picture below). I would like to install a Gigabyte GV-N250OC-1GI. 
There is a power supply and only 1 of the PCIe slots is in use and that would be

slot 1.  I’m pretty sure it will work, just not 100% sure. 
Also, once I’ve installed it, I’m not quite sure what needs to be done prior to turning on the PC, during booting, drivers???

Short answer: nothing.  Windows should detect the new adapter. Worst case is that Windows will use a "generic" device-driver, to give you 800-by-600 resolution. Then, download the manufacturer's device-driver,  and install it to get the "optimal" resolution that is compatible with your monitor.

you indicate if I understood well that you want to install the two graphic cards, together?

both at the same time this may be insufficient in power supply

Ok, so in this case it should be fine

but difficult to guarantee this
you have the drivers available from the link given so well this model
