If I use the backups from my older all-in-one desktop for my new desktop (all-in-one), cam I still use my older PC the exact same as I always have? It's for a business and I needed a 2nd computer for assistant.

There is no problem with the old or new system as long as the backups are only of personal files such as documents, images, music, etc.
Once you log into your Microsoft account one the new system, if you have been using OneDrive, then the app will synchronize your documents. You can log into the cloud to set what you want synchronized and put on the new system.
Note that his works two ways and you need to look at what you are going to share with your assistant and vice-versa..
When you log into your Microsoft account you can check your subscriptions and download any licensed software to the new PC
If using the Microsoft Windows backup tools you might want to ask Microsoft support.
