I have the OMEN 15.6 inch Gaming Laptop PC 15-en1000 (2L1F1AV) and I want to upgrade to 32 GB in only one slot just to keep one open for later on. Can I upgrade with DDR 4 3200 or do both slots need to be filled?

What is your OMEN 15.6 inch Gaming Laptop PC 15-en1000 (2L1F1AV) machine ? Machines of the series have 2 slots of RAM and support 32GB (2x16GB) DDR4 3200MHz RAM (from manual) or even 64GB using compatible RAM such as

Welcome to our HP Community Forum!
According to Crucial (see: https://www.crucial.com/compatible-upgrade-for/hp---compaq/hp-omen-15-en1000), you can upgrade your laptop with up to 2 x 32GB of DDR4 PC4-25600, 3200 MHz, Non-ECC, Unbuffered, SODIMM, 260-pin RAM sticks. And yes, you should be able to run your laptop with just one 32GB RAM stick. Please note that if you use only one RAM stick, your RAM won't run in either dual channel or dual rank mode.
So, why not buy two 32GB RAM sticks such as this purchase option: https://www.amazon.com/Timetec-Performance-PC4-25600-Unbuffered-Compatible/dp/B08KRXFM4R/ref=sr_1_4?... in order to achieve maximum RAM speed and performance, and not least because these RAM pairs happen to be priced very favorably these days, and be done with it.
