My HP Notebook 14-am054tx original wifi card has only 1 antenna. I like to upgrade the card to WiFi 6 card, should I need to add another antenna and should I need to route the antenna inside the LCD casing?

Do yourself a favor and save a lot of work and expense and do an easy swap to this dual band Wi-Fi 5 card that works with one antenna wire.
Realtek RTL8821CE 802.11 ac 1x1 Wi-Fi + Bluetooth 4.2 Combo Adapter (MU-MIMO supported) HP part # L17365-005.
Otherwise, you will need to find and buy the dual antenna kit listed in chapter 3, page 32 of your notebook, completely disassemble the notebook including the display panel and hinges to properly route the second antenna wire.

I found a Realtek RTL8821CE wifi card but the Part No. is different.
And, this is the original wifi card installed in my notebook. I've notice that the pins on the feet are different. Will this work in my HP Notebook?

The Realtek RTL8821CE wifi card even with one connector/antenna is dual band with 5ghz and 2.4ghz?
And is it ok to use the original single antenna installed in my Notebook? By retaining it, the new wifi card will be dual band wifi?

Yes, that is correct.
The notebook's RTL8821CE Wi-Fi card is dual band and both bands and Bluetooth will work with the single antenna wire your notebook currently has installed.
